目的考察新疆两色金鸡菊乙酸乙酯(EAE)和正丁醇(BE)提取物对自由基清除能力和醛糖还原酶的抑制活性。方法以叔丁基对羟基茴香醚(BHA)和维生素C(Vitamin C)为阳性对照,分别测定提取物对DPPH、ABTS、羟自由基(*OH)和超氧阴离子(*O_2~-)等自由基的清除能力;以槲皮素为阳性对照,DL-甘油醛为底物,醛糖还原酶(来源于大鼠眼晶状体)和辅酶NADPH组成的反应体系,测定提取物抑制醛糖还原酶的活性。结果与阳性对照BHA和Vitamin C比较,两色金鸡菊提取物对DPPH、ABTS、*OH和*O_2~-等自由基有不同程度的清除作用;与阳性对照药物槲皮素相比,提取物对醛糖还原酶的抑制能力用IC_(50)值表示,差异有统计学意义(P<0.05),提取物对醛糖还原酶有一定的抑制作用但弱于槲皮素。结论新疆两色金鸡菊乙酸乙酯和正丁醇提取物有清除自由基的能力,同时具有一定抑制醛糖还原酶的活性。
Objective To investigate the scavenging activity of free radicals and the inhibitory activity of aldose reductase (ACE) and n-butanol (BE) extracts of two species of Coreopsis. The extract had a certain inhibitory effect on aldose reductase but was weaker than quercetin. Conclusion The extract of ethyl acetate and n-butanol of Xinjiang bifidum chinense L. has the ability to scavenge free radicals and has certain inhibitory activity on aldose reductase.
