目的:利用大孔吸附树脂纯化新疆两色金鸡菊中的总黄酮。方法:比较10种大孔吸附树脂对响应面优化提取所获得的新疆两色金鸡菊总黄酮的吸附和解吸性能,考察不同类型的吸附树脂对总黄酮的吸附和解吸效果。结果:AB-8型大孔吸附树脂对新疆两色金鸡菊总黄酮具有较强的吸附性能和较好的解吸能力。当总黄酮上样浓度为0.54 mg/m L,上样量为树脂体积的2.1倍(2.1 BV),吸附平衡4 h后,先用2~3 BV蒸馏水冲洗柱床,再用2.2 BV的50%乙醇洗脱,分段收集,可获得较高含量的总黄酮有效部位。结论:AB-8型大孔吸附树脂可以作为纯化新疆两色金鸡菊总黄酮的材料。
Objective: To purify total flavonoids in two color Coreopsis chinensis using macroporous adsorption resin. Methods: The adsorption and desorption properties of total flavonoids from two species of Coreopsis chinensis collected by response surface optimization were compared by 10 macroporous adsorption resins. The adsorption and desorption effects of different types of adsorption resins on total flavonoids were investigated. Results: AB-8 macroporous resin had strong adsorption capacity and good desorption capacity for total flavonoids of two-color Coreopsis sp. In Xinjiang. When the concentration of total flavonoids was 0.54 mg / m L, the sample volume was 2.1 times (2.1 BV) of the resin volume. After 4 hours adsorption equilibrium, the column was washed with 2-3 BV distilled water, % Ethanol elution, segmented collection, to obtain a higher content of total flavonoids effective parts. Conclusion: The AB-8 macroporous resin can be used as a material for purification of flavonoids in Xinjiang bifidum.
