目的:探讨右归丸含药血清体外诱导大鼠骨髓基质干细胞分化为神经元样细胞的最佳浓度。方法:用DMEM/F-12培养液培养Wistar大鼠骨髓基质干细胞(BMSCs),采用不同浓度右归丸含药血清对其进行体外诱导,并用MTT法检测各组细胞生长情况,用免疫细胞荧光方法计算各组神经元样细胞阳性数加以比较。结果:BMSCs在2.5%含药血清培养液中活力最好。结论:2.5%右归丸含药血清培养液是诱导BMSCs分化实验的最佳浓度。
OBJECTIVE: To explore the optimal concentration of YGG containing serum in vitro to induce differentiation of rat bone marrow stromal stem cells into neuron-like cells. Methods: Wistar rat bone marrow stromal stem cells (BMSCs) were cultured in DMEM / F-12 medium and induced with different concentrations of Yiguowu pill serum in vitro. The cell growth was detected by MTT assay. Immunofluorescence Methods Calculate the positive number of neuron-like cells in each group to be compared. Results: BMSCs had the best activity in 2.5% serum-containing medium. Conclusion: 2.5% Yugui pills containing serum-containing medium is the best concentration for inducing BMSCs differentiation experiment.
