普洱茶茶褐素对大鼠激素敏感性脂肪酶活性及其mRNA表达的影响

目的研究普洱茶茶褐素对大鼠肝脏和附睾脂肪组织中HSL活性及其mRNA表达的影响。方法 90只SD大鼠随机分成正常对照组(Ⅰ组)、高脂饲料组(Ⅱ组)和高脂饲料+茶褐素组(Ⅲ组),每组选取10只于实验D15、D30和D45处死,分别测定其血清TG、TC、LDL-C和HDL-C值以及肝脏和附睾脂肪组织中HSL活性及其mRNA表达水平。结果 (1)试验D30、D45时,Ⅱ组大鼠TG、TC、LDL-C水平均较Ⅰ组显著升高(P<0.05,P<0.01),HDL-C水平较Ⅰ组显著降低(P<0.05,P<0.01),而Ⅲ组大鼠TG、TC、LDL-C水平则较Ⅱ组显著降低(P<0.05,P<0.01),HDL-C水平较Ⅱ组显著升高(P<0.05,P<0.01)。(2)D30时,Ⅲ组附睾脂肪组织HSL活性高于Ⅰ组(P<0.05);D45时,Ⅲ组肝脏组织HSL活性高于Ⅰ组(P<0.05),Ⅱ组和Ⅲ组中附睾脂肪组织HSL活性显著高于Ⅰ组(P<0.01)。(3)D30时,Ⅱ组和Ⅲ组中附睾脂肪组织HSLmRNA表达则分别高于Ⅰ组(P<0.05,P<0.01),且Ⅱ组和Ⅲ组之间差异极显著(P<0.01);D45时,Ⅲ组大鼠肝脏组织HSL mRNA表达明显高于Ⅰ组(P<0.05),Ⅱ组和Ⅲ组中附睾脂肪组织HSLmRNA表达则均高于Ⅰ组(P<0.05,P<0.01),且Ⅱ组和Ⅲ组之间差异极显著(P<0.01)。结论普洱茶茶褐素对大鼠降血脂效果明显,并可显著增强大鼠肝脏和附睾脂肪组织HSL活性及其mRNA的表达,是茶褐素降血脂功效的重要作用机制之一。 Objective To investigate the effect of Pu-erh tea-chaff on HSL activity and mRNA expression in rat liver and epididymis adipose tissue. Methods Totally 90 SD rats were randomly divided into normal control group (group Ⅰ), high fat diet group (group Ⅱ) and high fat diet + The serum levels of TG, TC, LDL-C, HDL-C and the activities of HSL in the liver and epididymis adipose tissue were determined. Results The levels of TG, TC and LDL-C in group Ⅱ were significantly higher than those in group Ⅰ (P <0.05, P <0.01) and HDL-C in group Ⅱ were significantly lower than those in group Ⅰ (P <0.05, P <0.01), but TG, TC and LDL-C in group Ⅲ were significantly lower than those in group Ⅱ (P <0.05, P <0.01) 0.05, P <0.01). (2) The HSL activity of epididymal adipose tissue of group Ⅲ was higher than that of group Ⅰ at D30 (P <0.05), while the activity of HSL of group Ⅲ was higher than that of group Ⅰ at D45 (P <0.05) Tissue HSL activity was significantly higher than the group Ⅰ (P <0.01). (3) At D30, the expression of HSLmRNA in epididymal adipose tissue was higher in group Ⅱ and group Ⅲ than that in group Ⅰ (P <0.05, P <0.01), and the difference between group Ⅱ and group Ⅲ was significant (P <0.01). (P <0.05). HSL mRNA expression in epididymal adipose tissue was higher in group Ⅱ and group Ⅲ than that in group Ⅰ (P <0.05, P <0.01) There was significant difference between group Ⅱ and group Ⅲ (P <0.01). Conclusions Pu-erh tea-scented tea extract has a significant effect on reducing blood fat in rats, and can significantly enhance HSL activity and mRNA expression in rat liver and epididymis adipose tissue. It is one of the important mechanisms of theabrownin’s hypolipidemic effect.
