  ATP is currently regarded as the cell death factor not only the energy.We have found previously that hydrogen sulphide (H2S) is protective against ischaemia-reperfusion injury of the brain.But the molecular mechanisms are still unclear.In the current study, we investigated the cytotoxicity of ATP (1, 3, 5, 7, 9 mM) in neuronal PC12 cells by the morphology and XTT assay.The change of H2S/cystathionine beta-synthase (CBS)in PC12 cells damaged by ATP were detected by immunofluorescence and western blotting.Further we observed the effect of endogenous and exogenous H2S on PC12 cells respectively treatmented with the CBS inhibitor,aminooxyacetic acid (AOAA).and NaHS (the donor of exogenous H2S).We found PC12 cells became edema,the cellular pustutes were thinner and shorter after exposure to ATP Cell vitality decreased in a manner of time dependence.The content of H2S and CBS increased in PC12 cells injuryed by ATP AOAA enhanced the injury-induced effect by ATP and NaHS attenuated the injury induced by ATP Finally we concluded that higher concentration of ATP induced the injury in PC12 cells, and H2S could protect the cell injury induced by ATP.
