The change of brain metabolism and the action of aspirin after cerebral ischemia-reperfusion injury

  This study investigated and compared the change of brain metabolis and the action of aspirin after cerebral ischemia-reperfusion injury (CIRI).Right middle cerebral artery was occluded by inserting a thread through internal carotid artery for 2 h, and then reperfused for 24 h or 72 h.60 mg/kg dose of aspirin was intragastric administrated at reperfusion 0 h.The brain injured area, and the neuronal densitywere estimated.The apoptosis determined by TUNEL method.The lactate (LA) content, lactate dehydrogenase (LDH), and Na+-K+-ATPase were measured by biochemical methods.The content of adenosion5-triphosphate (ATP) in brain tissue was separated by HPLC.After CIRI 24 h, the brain injured area, and the number of apoptosis were significantly increased.The neuronal density in injured regions, the content of LA and ATP, the activity of LDH and Na+-K+-ATPase were decreased.Compare with CIRI 24 h, the brain injured area, the neuron density, and the number of apoptotic cells was improved, the content of LA and ATP, the activity of Na+-K+-ATPase was reduced furtherly, and the activity of LDH was increased on CIRI rats for 72 h.With using of 60 mg/kg dose of aspirin, all of the index were belance to normal for CIRI 24 h or 72 h.
