目的探讨陈皮半夏是否能通过衰老脐静脉内皮细胞(HUVEC)中磷脂酰肌醇3激酶-蛋白激酶B(PI3K-Akt)信号通路对p53和p21的表达进行干预。方法半夏、陈皮按照1∶1的比例配伍,常法煎煮制备陈皮半夏汤剂。对5只SD大鼠使用陈皮半夏汤剂灌胃,分离血清。使用0.25%胰岛素-柠檬酸盐诱导HUVEC衰老,使用流式细胞仪检测细胞周期,建立细胞衰老模型后,通过Western blot法检测陈皮半夏干预衰老HUVEC中PI3K、磷酸化Akt(p-Akt)、p53和p21的表达。结果 0.25%胰岛素-柠檬酸盐诱导可以有效建立细胞衰老模型。与空白对照组和陈皮半夏对照组比较,模型组中PI3K、p-Akt、p53和p21表达显著升高(P<0.01);陈皮半夏和PI3K特异性阻滞剂LY294002干预后,PI3K、p-Akt、p53和p21表达较模型组均显著降低(P<0.05)。结论陈皮半夏有可能通过衰老HUVEC中PI3K-Akt通路调控p53和p21的表达。
Objective To investigate whether the dried orange peel can intervene the expression of p53 and p21 through the PI3K-Akt signaling pathway in senescent human umbilical vein endothelial cells (HUVECs). Methods Pinellia, dried tangerine peel in accordance with the ratio of 1: 1 compatibility, often decoction prepared dried orange peel Decoction. 5 SD rats using dried tangerine peel soup gavage, serum separation. The senescence of HUVECs was induced by 0.25% insulin-citrate. The cell cycle was detected by flow cytometry. After establishment of cellular senescence model, the effects of dried tangerine peel on the expression of PI3K, p-Akt, p53 and p21 expression. Results 0.25% insulin-citrate induction could effectively establish cellular senescence model. PI3K, p-Akt, p53 and p21 expression in the model group were significantly increased compared with the blank control group and the dried orange peel control group (P <0.01). After the intervention of PYP and PI3K-specific blocker LY294002, the expressions of PI3K, The expression of p-Akt, p53 and p21 were significantly lower than the model group (P <0.05). Conclusion The dried tangerine peel is likely to regulate the expression of p53 and p21 through the PI3K-Akt pathway in senescent HUVECs.
