目的研究氟致大鼠睾丸组织的损伤作用以及钙的拮抗作用。方法选用健康雄性清洁级SD大鼠80只,随机分为10组:对照组、加钙染氟6个剂量组(NaF 2.1、4.2、8.5、17、34、68 mg/kg·d;CaCO325、50、50、100、100、150 mg/kg·d)和单纯染氟3个剂量组(NaF 2.1、8.5、34 mg/kg·d),灌胃染毒90 d,染毒结束次日,观察氟、钙对大鼠睾丸脏器系数、睾丸结构的影响,并用氟离子选择电极法检测睾丸氟含量,用原子吸收分光光度计测定睾丸中钙(Ca)、铁(Fe)和锌(Zn)元素水平。结果氟可使大鼠睾丸脏器系数降低并导致睾丸病理学改变。各组大鼠睾丸氟含量组间比较,差异有统计学意义(F=62.224,P=0.000)。表现为加钙染氟第4、5、6组和高、中、低剂量染氟组睾丸氟含量均高于对照组。高、中剂量染氟浓度下加钙组睾丸氟含量低于单纯染氟组。各组大鼠睾丸Ca、Fe和Zn水平组间比较,差异有统计学意义(F值分别为7.658、6.177、10.802,P<0.05),表现为高、中、低剂量染氟组Ca、Fe和Zn水平均低于对照组,加钙组睾丸Ca、Fe和Zn水平高于高剂量染氟组。结论氟可在睾丸内蓄积、破坏睾丸结构并改变睾丸中一些金属元素水平,适量的钙可拮抗氟致大鼠睾丸组织的损害作用。
Objective To study the injury of fluoride-induced testicular tissue and the antagonism of calcium. Methods Eighty healthy and male SD rats were randomly divided into 10 groups: control group, 6 doses of calcium fluoride (NaF 2.1, 4.2, 8.5, 17, 34 and 68 mg / kg · d; CaCO325, 50, 50, 100, 100 and 150 mg / kg · d) and 3 groups (NaF 2.1, 8.5 and 34 mg / kg · d) respectively. The effect of fluoride and calcium on testis organ coefficient and testicular structure were observed. Fluoride content was measured by fluoride ion selective electrode method. Results Fluoride can reduce the testis organ coefficient in rats and lead to pathological changes of testis. There was significant difference between the testicular fluorine levels in each group (F = 62.224, P = 0.000). Fluoride group showed higher levels of fluorine in testicles than those in control group in groups 4, 5, and 6 with high calcium, medium and low doses of fluorine. High and medium doses of fluoride concentration in the test group with calcium fluoride test group fluoride content was lower than the simple fluorine group. The levels of Ca, Fe and Zn in testis of rats in each group were significantly different (F = 7.658, 6.177, 10.802, respectively, P <0.05) And Zn levels were lower than the control group, Ca, Fe and Zn levels in testicular calcium group were higher than those in high-dose fluoride group. Conclusion Fluorine can accumulate in testis, destroy the structure of testis and change the level of some metal elements in testes. The proper amount of calcium can antagonize the damaging effects of fluoride on testicular tissue in rats.
