采用40%丙酮提取灰树花子实体中的多酚类物质,经NKA-Ⅱ型大孔吸附树脂纯化,然后依次用乙酸乙酯、正丁醇萃取制备不同极性的多酚组分。通过分析总还原力、DPPH自由基清除率、羟自由基清除率、亚铁离子螯合能力,从而研究不同极性多酚组分的抗氧化活性。结果表明,不同多酚质量浓度的乙酸乙酯相总还原力均高于正丁醇相和水相;乙酸乙酯相的DPPH自由基清除率IC50值为12.192μg/m L,且羟自由基清除率IC50值为66.040μg/m L,说明乙酸乙酯萃取相中所含的多酚组分抗氧化活性较强。
Polyphenols from Grifola frondosa were extracted with 40% acetone, purified by NKA-Ⅱ macroporous adsorption resin, and then extracted with ethyl acetate and n-butanol to prepare polyphenol fractions with different polarities. By analyzing the total reducing power, DPPH free radical scavenging rate, hydroxyl radical scavenging rate and ferrous ion chelating ability, the antioxidant activity of polyphenols with different polarities was studied. The results showed that the total reducibility of ethyl acetate phase with different concentrations of polyphenol was higher than that of n-butanol phase and water phase. The IC50 value of DPPH radical scavenging activity of ethyl acetate phase was 12.192μg / m L, The IC50value of clearance was 66.040μg / m L, indicating that the polyphenol component contained in the ethyl acetate extract phase had stronger antioxidant activity.
