目的探讨硫化氢(H_2S)对不同周龄自发性高血压大鼠(SHR)离体脑基底动脉舒张反应的影响。方法选取14、40周龄SHR和正常Wistar京都(WKY)大鼠各6只,测量血压后,急性分离脑基底动脉,应用压力肌动图技术检测不同浓度硫氢化钠(NaHS)对大鼠脑基底动脉直径变化的影响。结果 (1)10、30、100、300、1 000、3 000 mmol/L的NaHS能够浓度依赖地舒张SHR和WKY大鼠脑基底动脉;(2)与14周龄的WKY相比,1 000、3 000 mmol/L的Na HS对14周龄SHR的舒张率更明显,两者间有显著性差异(P<0.05);(3)与40周龄的WKY大鼠相比,1 000、3 000 mol/L的NaHS对40周龄SHR的舒张率降低,两者间有显著性差异(P<0.05)。结论在高血压初期,脑基底动脉对H_2S的敏感性增加,而随着高血压的发展及血管功能的严重损伤,脑基底动脉可能对H_2S的敏感性降低。
Objective To investigate the effect of hydrogen sulfide (H2S) on the relaxation of isolated basilar artery in spontaneously hypertensive rats (SHR) of different weeks. Methods Six SHRs of 14, 40 weeks old and WKY rats (Wistar rats) were randomly divided into 6 groups. After blood pressure was measured, basilar artery was isolated acutely and pressuredrogram was used to detect the effects of sodium hydrosulfide (NaHS) The influence of basilar artery diameter changes. Results (1) NaHS at 10, 30, 100, 300, 1000 and 3000 mmol / L could relax the basilar artery of SHR and WKY rats in a concentration-dependent manner. (2) Compared with WKY at 14 weeks, 1 000 (P <0.05). (3) Compared with 40-week-old WKY rats, 1 000, The relaxation rate of NaHS at 3 000 mol / L decreased at 40 weeks of age, with significant difference (P <0.05). Conclusions In the early stage of hypertension, the sensitivity of cerebral basilar artery to H 2 S increased. However, with the development of hypertension and the severe damage of vascular function, the sensitivity of cerebral basilar artery to H 2 S may be decreased.
