目的 观察诱导分化剂二甲基甲酰胺(DMF)对胶质瘤细胞的增殖抑制作用及DMF对癌基因C-myc蛋白水平表达的影响。方法 [3H]-TdR掺入法测C6细胞增殖率;免疫组化SP法测C-myc基因蛋白水平的表达。结果 应用二甲基甲酰胺后,C6胶质瘤细胞CPM值降低,DNA合成总量减少。C-myc基因胞浆阳性表达下降。结论 二甲基甲酰胺对胶质瘤细胞存在剂量依赖性抑制。作用机理与DMF降低了癌基因C-myc对C6细胞恶性增殖的转录调节作用有关。
Objective To observe the effect of dimethyl formamide (DMF) on the proliferation of glioma cells and the effect of DMF on the expression of oncogene C-myc. Methods The [3H] -TdR incorporation assay was used to determine the proliferation rate of C6 cells. The expression of C-myc protein was detected by immunohistochemical SP method. Results After using dimethylformamide, the C6 glioma cell CPM value decreased, the total amount of DNA synthesis decreased. Cytoplasm positive expression of C-myc gene decreased. Conclusion Dimethylformamide inhibits glioma cells in a dose-dependent manner. The mechanism of action and DMF reduce the oncogene C-myc on C6 cell proliferation and transcriptional regulation.
