目的研究慢性丙型肝炎病毒(HCV)感染患者外周血CD81、CD4的表达,探讨CD81对患者机体免疫系统的调节作用。方法选择HCV感染患者55例(感染组),健康对照25例(健康对照组),采用流式细胞术检测其外周血CD81、CD4的表达水平,ELISA法检测血清中白介素2(IL-2)的含量,RT-PCR法检测血清中HCVRNA的含量。结果感染组CD81分子的表达水平与血清中HCVRNA的含量呈正相关(r=0.474,P<0.01)。感染组外周血淋巴细胞中CD81+细胞和CD3+CD4+T细胞中CD3+CD4+CD81+T细胞的数量均高于健康对照组(P<0.01),且二者呈正相关(r=0.581,P<0.01);感染组CD3+CD4+T细胞的数量和血清中IL-2的含量均显著低于健康对照组(P<0.05),且两者呈正相关(r=0.378,P<0.05)。结论 HCV感染后,HCVRNA的持续存在可能促进细胞表面CD81分子的表达;CD81分子高表达的同时,伴随CD3+CD4+T细胞数量降低,且分泌的细胞因子IL-2减少,参与患者机体的免疫调节。
Objective To investigate the expression of CD81 and CD4 in peripheral blood of patients with chronic hepatitis C virus (HCV) infection and to explore the regulatory effect of CD81 on the immune system of patients. Methods 55 cases of HCV infection (infected group) and 25 healthy controls (healthy control group) were selected. The expression of CD81 and CD4 in peripheral blood was detected by flow cytometry. The levels of interleukin 2 (IL-2) The content of HCV RNA in serum was detected by RT-PCR. Results The expression of CD81 in the infected group was positively correlated with the level of HCV RNA in serum (r = 0.474, P <0.01). The number of CD3 + CD4 + CD81 + T cells in CD81 + cells and CD3 + CD4 + T cells in peripheral blood lymphocytes in infected group was significantly higher than that in healthy controls (P <0.01), and there was a positive correlation between them (r = 0.581, P <0.01). The number of CD3 + CD4 + T cells and the level of IL-2 in serum of infected patients were significantly lower than those of healthy controls (P <0.05), and there was a positive correlation between them (r = 0.378, P <0.05). CONCLUSIONS: The persistence of HCV RNA may promote the expression of CD81 on the cell surface after HCV infection. CD81 molecules are highly expressed, accompanied by the decrease of the number of CD3 + CD4 + T cells and the decrease of secreted cytokine IL-2 adjust.
