目的:探讨B71单独或者与IL6联合在体外诱导T细胞活化的能力。方法:在混合淋巴细胞肿瘤细胞培养(MLTCs)后,测定淋巴细胞增殖指数和CTL杀伤活性。结果:B71+的B16细胞能够较有效地刺激淋巴细胞增殖、诱导特异CTL杀伤活性(与野生型B16细胞和模拟转染的B16细胞比较,P<0.01),当与IL6结合时效果更加显著(与各对照组比较P<0.01)。结论:B71基因在肿瘤细胞中表达可增强其免疫原性,在体外有效诱导T细胞活化;在此过程中IL6与B71分子存在协同效应。
AIM: To investigate the ability of B71 alone or in combination with IL6 to induce T cell activation in vitro. Methods: After mixed lymphocyte tumor cell culture (MLTCs), the lymphocyte proliferation index and CTL killing activity were determined. RESULTS: B71 + B16 cells stimulated lymphocyte proliferation more efficiently and induced specific cytotoxicity against CTLs (P <0.01 compared with wild-type B16 cells and mock-transfected B16 cells) and more significantly when combined with IL6 (and Each control group P <0.01). CONCLUSION: The expression of B71 gene in tumor cells enhances its immunogenicity and effectively induces T cell activation in vitro. Synergistic effects of IL-6 and B71 exist in this process.
