目的 探讨补锌对衰老过程睾丸细胞凋亡的影响及其可能机制。方法 选用 2 4月龄 Wistar雄性大鼠 1 6只 ,均分为实验组和对照组 ,实验 (补锌 )组和对照组分别喂养补锌和常锌饲料。喂养 4 w后 ,采用原子吸收分光光度法测定血清锌含量 ;采用放射免疫分析法测定血清睾丸酮含量 ;采用 NADPH- d组织化学法显示睾丸细胞一氧化氮合酶 (NOS)活性 ;采用原位缺口平移技术 (INST法 )显示睾丸凋亡细胞数目。结果 与对照组相比 ,老年大鼠补锌 4w后 ,血清锌含量明显升高 (P<0 .0 5) ;血清睾丸酮水平明显升高 (P<0 .0 5) ;睾丸间质细胞 NOS阳性细胞数目明显减少 (P<0 .0 1 ) ,NOS阳性细胞反应强度明显减低 ;睾丸生精细胞凋亡数目明显减少 (P<0 .0 5)。结论 锌是睾丸发育必需的营养素 ,补锌可使老年大鼠睾丸细胞凋亡数目减少 ,这种变化可能与补锌对衰老大鼠睾丸细胞 NOS活性和睾丸酮水平的调节有关。
Objective To investigate the effect of zinc supplementation on testicular cell apoptosis in aging and its possible mechanism. Methods Twenty-six male Wistar rats aged 24 months were divided into experimental group and control group. The experimental group (zinc supplement) and the control group were fed zinc supplementation and zinc supplementation respectively. Serum zinc levels were measured by atomic absorption spectrophotometry after feeding for 4 weeks. Serum testosterone levels were determined by radioimmunoassay. Nitric oxide synthase (NOS) activity in testis cells was detected by NADPH-d histochemistry. The translation technique (INST method) shows the number of testicular apoptotic cells. Results Compared with the control group, the levels of serum zinc increased significantly (P <0.05) in serum and serum testosterone levels (P <0.05) in the aged rats after zinc supplementation for 4 weeks. The levels of NOS The number of positive cells decreased significantly (P <0.01), and the reaction intensity of NOS positive cells decreased significantly. The number of apoptotic spermatogenic cells in testis decreased significantly (P <0.05). Conclusion Zinc is an essential nutrient for testicular development. Zinc supplementation can reduce the number of apoptosis of testicular cells in aged rats, which may be related to the regulation of NOS activity and testosterone level in testicular cells of aged rats by zinc supplementation.
