目的 :探讨p16基因在体内对恶性胶质瘤的生长抑制作用。方法 :将外源性p16基因导入C6 细胞内 ,应用立体定向技术将C6细胞种植于SD大鼠尾状核头部 ,用核磁共振 (MR)扫描技术 ,动态观察颅内肿瘤生长情况。并通过免疫组化、原位杂交和细胞凋亡检测肿瘤细胞的增殖活性。结果 :转染组和治疗组大鼠生存期较对照组明显延长。治疗组肿瘤随时间的延长逐渐缩小。免疫组化显示转染组和治疗组P16蛋白表达明显增强。原位杂交和细胞凋亡检测表明 ,转染组和治疗组大鼠肿瘤细胞增殖活性降低。结论 :p16基因在体内有抑制恶性胶质瘤生长的作用 ;瘤体内注入p16cDNA质粒 脂质体复合物 ,可使肿瘤生长受到明显抑制 ,并使多数肿瘤消失
Objective: To investigate the inhibitory effect of p16 gene on the growth of malignant glioma in vivo. Methods: Exogenous p16 gene was introduced into C6 cells. C6 cells were seeded into the caudate nucleus of SD rats by stereotactic technique. The growth of intracranial tumors was observed by using magnetic resonance (MR) technique. The proliferative activity of tumor cells was detected by immunohistochemistry, in situ hybridization and apoptosis. Results: The survival of rats in the transfection group and the treatment group was significantly longer than that in the control group. The treatment group tumors gradually reduced over time. Immunohistochemistry showed that transfection group and treatment group P16 protein expression was significantly enhanced. In situ hybridization and apoptosis detection showed that the transfected group and treated group rats tumor cell proliferation activity decreased. Conclusion: The p16 gene can inhibit the growth of malignant glioma in vivo. The injection of p16 cDNA plasmid liposome complex in the tumor can significantly inhibit the growth of the tumor and eliminate the majority of tumors
