目的:观察葛根素对链脲佐菌素诱导2型糖尿病大鼠的膀胱逼尿肌形态学的影响。方法:雌性大鼠分为正常对照组、糖尿病组、糖尿病葛根素治疗组。以高糖高脂饮食及小剂量链脲佐菌素(30 mg/kg)制备2型糖尿病大鼠模型。在模型成功的Ⅱ型糖尿病大鼠中随机再分为糖尿病无干预组(12只),16周后制备逼尿肌标本,在光镜下观察膀胱逼尿肌在常规HE染色、VanGieson氏苦味酸酸性品红染色膀胱黏膜、固有层及膀胱肌层改变,和石蜡切片明胶镀银染色观察神经纤维的改变。结果:16周后葛根素治疗组大鼠的血糖值、膀胱湿重、膀胱相对重量比糖尿病组显著改善,苦味酸酸性品红(V-G)染色膀胱逼尿肌间胶原绝对含量葛根素治疗组较糖尿病组明显下降。结论:在链脲佐菌素诱导糖尿病大鼠葛根素具有保护膀胱逼尿肌功能。
Objective: To observe the effects of puerarin on the detrusor muscle morphology of streptozotocin-induced type 2 diabetic rats. Methods: Female rats were divided into normal control group, diabetic group and diabetic puerarin treatment group. Type 2 diabetic rat model was induced by high-fat and high-fat diet and low-dose streptozotocin (30 mg / kg). Twelve diabetic rats were randomly divided into two groups: diabetic rats without intervention group (n = 12), and detrusor muscle specimens were prepared after 16 weeks. The detrusor of the bladder was observed under light microscope by HE staining and van Gieson’s picric acid Acid magenta staining of bladder mucosa, lamina propria and bladder muscle layer changes, and paraffin sections of silver staining of gelatin to observe changes in nerve fibers. Results: After 16 weeks, the blood glucose, bladder wet weight and bladder relative weight of puerarin-treated group were significantly improved than those of diabetic group. The content of collagen in detrusor muscle of VG group was significantly lower than that of puerarin-treated group Diabetes group decreased significantly. Conclusion: puerarin in streptozotocin-induced diabetic rats protects detrusor function of the bladder.
