目的探讨IL-10启动子基因-592(C/A)多态性与泸州市汉族人群平均血糖、血脂水平的关系。方法采用PCR-RFLP分析方法结合DNA直接测序法对258例正常人群、196例2型糖尿病人群及448例血脂异常人群进行IL-10-592(C/A)多态性分析。结果正常人群中AA基因型HDL-C水平较CC+AC基因型低,且差异有统计学意义(P<0.05),其他指标差异无统计学意义。2型糖尿病人群血糖各项指标在基因型间无明显差异,血脂异常人群血脂各项指标在基因型间无明显差异,但低HDL-C人群中基因型AA与基因型CC+AC者相比,HDL-C水平降低,TC和LDL-C水平增高,且差异有统计学意义(P<0.05)。结论 IL-10启动子基因-592(C/A)多态性与泸州市人群平均血脂水平相关,AA基因型可能引起HDL-C水平的降低。
Objective To investigate the relationship between the -592 (C / A) polymorphism of IL-10 promoter and the average blood glucose and serum lipids in Han nationality in Luzhou. Methods Polymorphisms of IL-10-592 (C / A) in 258 normal subjects, 196 patients with type 2 diabetes mellitus and 448 patients with dyslipidemia were analyzed by PCR-RFLP and DNA sequencing. Results Compared with CC + AC genotype, HDL-C in AA genotype was lower in normal subjects (P <0.05). There was no significant difference in other indexes. There was no significant difference among the genotypes in blood glucose among all the type 2 diabetic patients. There was no significant difference in the blood lipid among all the subjects with type 2 diabetes mellitus, but genotypes AA and CC + AC in low HDL-C , HDL-C levels decreased, TC and LDL-C levels increased, and the difference was statistically significant (P <0.05). Conclusion The -592 (C / A) polymorphism of IL-10 promoter is associated with the average blood lipid level in Luzhou. AA genotype may reduce the level of HDL-C.
