目的探讨参芎葡萄糖注射液对小鼠急性心肌缺血再灌注(I/R)损伤的保护作用及可能的分子机制。方法建立急性心肌缺血再灌注(Ischemia-Reperfusion,I/R)损伤的小鼠模型,将野生型小鼠(C57BL/6)随机分为3组,假手术组(Sham),缺血再灌注+生理盐水组(Vehicle),缺血再灌注+参芎葡萄糖注射液预处理组(20mL/kg,SL),术前2小时给予腹腔注射生理盐水或参芎葡萄糖注射液,于缺血再灌注后不同时间点,通过心脏超声观察各组小鼠心功能的变化,心肌组织TTC染色确定心梗面积大小,脱氧核糖核苷酸末端转移酶法(TUNEL)检测凋亡心肌,蛋白免疫印迹法(Western Blot)及酶联免疫吸附法(ELISA)分别检测心肌组织以及血清炎症因子肿瘤坏死因子(TNF-α)表达水平变化。结果 I/R手术48小时后,与假手术组相比,左室射血分数(LVEF)明显降低(P<0.01)。参芎葡萄糖注射液预处理明显改善I/R组的LVEF,并降低了心肌梗死面积,抑制I/R引起的心肌细胞凋亡和组织炎症反应。进一步的研究表明,参芎葡萄糖注射液预处理明显抑制了I/R后心肌组织以及血清中TNF-α表达水平升高(P<0.01)。结论参芎葡萄糖注射液预处理通过抑制TNF-α表达水平发挥对缺血再灌注(I/R)损伤心肌的保护作用。
Objective To investigate the protective effect of Shenxiong glucose injection on acute myocardial ischemia-reperfusion (I / R) injury in mice and its possible molecular mechanism. Methods The mouse model of acute myocardial ischemia / reperfusion (I / R) injury was established. The wild type mice (C57BL / 6) were randomly divided into three groups: sham operation group, ischemia reperfusion + Vehicle, ischemia / reperfusion + Shenxiong glucose injection (20mL / kg, SL), intraperitoneal injection of normal saline or Shenxiong glucose injection 2 hours before operation, ischemia / reperfusion At different time points, the changes of cardiac function in each group were observed by echocardiography, myocardial infarct size was determined by TTC staining, apoptotic myocardium was detected by TUNEL, western blotting Western Blot and enzyme-linked immunosorbent assay (ELISA) were used to detect the expression of TNF-α in myocardial tissue and serum inflammatory cytokine. Results After 48 hours of I / R surgery, left ventricular ejection fraction (LVEF) was significantly lower than that in sham operation group (P <0.01). Pretreatment with Shenxiong and dextrose injection significantly improved LVEF in I / R group, reduced infarct size and inhibited cardiomyocyte apoptosis and tissue inflammatory response induced by I / R. Further studies showed that Shenxiong glucose injection pretreatment significantly inhibited myocardial I / R and serum TNF-α expression increased (P <0.01). Conclusion Pretreatment with Shenxiong and dextrose injection can exert a protective effect on myocardial injury induced by ischemia-reperfusion (I / R) injury by inhibiting the expression of TNF-α.
