目的:观察RalA和RhoC基因在非小细胞肺癌组织细胞中的表达变化,探讨它们的表达在非小细胞肺癌发展和侵袭转移中的作用。方法:通过实时荧光定量PCR(FQ-PCR)检测RalA、RhoC在60例非小细胞肺癌患者的肿瘤组织和癌旁组织细胞中的表达,了解其与非小细胞肺癌临床病理学指标的关系。结果:RhoC在非小细胞肺癌组织细胞中的表达较癌旁组织显著增高(P<0.05),且与淋巴结的转移、TNM分期和分化程度密切相关(P<0.05)。而RalA在非小细胞肺癌组织细胞中的表达量低于癌旁组织(P<0.05),与淋巴结转移无明显相关(P>0.05)。二者在鳞、腺癌中表达均无差异(P<0.05)。结论:肺癌组织细胞中RhoC表达升高及RalA表达异常下调与非小细胞肺癌的发生、发展及侵袭机制关系密切。
OBJECTIVE: To observe the changes of RalA and RhoC gene expression in non-small cell lung cancer cells, and to explore the role of RalA and RhoC in the development, invasion and metastasis of non-small cell lung cancer. Methods: The expression of RalA and RhoC in 60 cases of non-small cell lung cancer and adjacent non-small cell lung cancer tissues were detected by real-time fluorescence quantitative PCR (FQ-PCR), and their relationship with the clinicopathological parameters of non-small cell lung cancer was analyzed. Results: The expression of RhoC in non-small cell lung cancer tissues was significantly higher than that in adjacent non-small cell lung cancer tissues (P <0.05), and closely correlated with lymph node metastasis, TNM stage and differentiation (P <0.05). However, the expression of RalA in non-small cell lung cancer cells was lower than that in paracancer tissues (P <0.05), but not with lymph node metastasis (P> 0.05). There was no difference between the two in squamous cell carcinoma and adenocarcinoma (P <0.05). Conclusion: RhoC expression in lung cancer cells and abnormal expression of RalA are closely related to the occurrence, development and invasion of non-small cell lung cancer.
