以小麦基因型CB037、新春9号和Bobwhite为材料进行成熟胚和幼胚培养,研究了CuSO4和Fe2-EDTA对小麦组织培养再生效果的影响。结果发现,培养基中添加0.1~4.1μmol/L CuSO4对小麦成熟胚组织培养无显著效果,超过6.1μmol/L对愈伤组织有毒害作用;培养基中添加40μmol/L Fe盐对小麦成熟胚组织培养和植株再生具有促进作用;不同小麦基因型其幼胚培养对Fe盐的反应不同,40μmol/L~50μmol/L Fe盐处理对CB037幼胚再生效果最好,20μmol/L Fe盐浓度对新春9号和Bobwhite幼胚再生效果较好,高浓度铁盐抑制作用显著;在相同培养条件下,CB037幼胚再生率最高,新春9号次之,Bob-white最低。
The effects of CuSO4 and Fe2-EDTA on the regeneration of wheat tissue were studied by using wheat genotypes CB037, Xinchun 9 and Bobwhite as materials. The results showed that adding 0.1-4.1μmol / L CuSO4 to the culture medium had no significant effect on the tissue culture of wheat mature embryos, and more than 6.1μmol / L had toxic effects on the callus. Adding 40μmol / L Fe salt to the medium, Tissue culture and plant regeneration had a promoting effect. The response of different immature embryos to different Fe wheat genotypes differed from each other. The effects of 40μmol / L ~ 50μmol / L Fe salt treatment on regeneration of CB037 immature embryos were best, and 20μmol / L Fe salt concentration The regeneration of the young embryos of New Year 9 and Bobwhite was better, and the inhibitory effect of high concentrations of iron was significant. Under the same culture conditions, the regeneration rate of CB037 was the highest, followed by that of New Year 9, and Bob-white was the lowest.
