采用栓线法和四动脉结扎法造成大鼠脑缺血再灌注模型 ,观察了海洋硫酸多糖类药物“989”对缺血脑组织的保护作用。结果发现 ,“989”在 2~ 10 mg·kg-1范围内静脉注射 ,能明显缩小脑栓塞大鼠 2 4 h的脑梗塞范围 ,改善行为障碍 ,对局灶性脑缺血和全脑缺血均有明显治疗作用。进一步研究发现 ,其作用机制可能与抑制脑组织 TNF- α表达有关。
The cerebral ischemia-reperfusion model was induced by the method of plug and four-artery ligation in rats. The protective effect of marine sulfate polysaccharides “989” on ischemic brain tissue was observed. The results showed that intravenous administration of “989” in the range of 2 ~ 10 mg · kg-1 could significantly reduce the range of cerebral infarction in 24 h after cerebral embolism and improve behavioral disorders. Blood have obvious therapeutic effect. Further study found that the mechanism of action may be related to the inhibition of TNF-αexpression in brain tissue.
