我国猕猴群中B病毒分离鉴定和检测方法研究初报

本实验从野外捕获的恒河猴的口腔溃疡灶中分离到一株病毒。该病毒在Vero细胞上具有B病毒特征性细胞病变 ,电镜观察具有疱疹病毒的典型形态与结构。经PCR扩增、SacⅡ酶切、PCR产物测序并与美国分离的BVE2 4 90株的部分基因序列进行比较 ,初步证实该分离株为B病毒 ,并命名为BV1 4 7株。用BV1 4 7株制备抗原片 ,并与国产HSV 1抗原片同时进行猴血清中B病毒抗体检测 ,可明显提高阳性检出率。B病毒的分离成功 ,对于提高我国实验猕猴的质量和检测水平 ,建立无B病毒猴群 ,增加出口创汇均具有十分重要的意义。 In this experiment, a virus was isolated from the oral ulcers of the rhesus monkeys captured in the wild. The virus has V cells characteristic of B virus disease, electron microscopy with herpes virus typical morphology and structure. The results of PCR amplification, SacⅡ digestion, sequencing of PCR products and comparison with partial sequences of BVE2 4 90 strains in the United States showed that the isolate was B virus and named BV1 47 strains. The BV1 47 strain was used to prepare the antigenic fragment, and simultaneous detection of the B virus antibody in the monkey serum with the domestic HSV1 antigenic fragment could obviously improve the positive detection rate. The success of the isolation of B virus is of great significance for improving the quality and testing level of experimental macaques in our country, establishing a group of monkeys without B virus and increasing export earnings.
