目的:探讨人肿瘤坏死因子α(TNF-α)联合紫杉醇对人腺样囊性癌细胞系SACC-83的抑制增殖作用及与细胞凋亡的关系,为化疗与生物治疗联合应用治疗腺样囊性癌提供实验依据。方法:将人腺样囊性癌细胞系SACC-83和对照组原代培养鼠肺成纤维细胞根据不同药物浓度分为9组,药物作用72 h。3H-TDR法检测细胞增殖抑制率;流式细胞术检测细胞凋亡率及细胞周期改变;透射电镜观察凋亡细胞形态变化。结果:联合用药组的细胞增殖抑制率显著高于单独用药组(P<0.05)。L929细胞各组的细胞增殖抑制率显著低于SACC-83细胞(P<0.05)。结论:TNF-α联合紫杉醇对SACC-83细胞增殖的抑制作用以及诱导凋亡具有协同作用。
Objective: To investigate the relationship between tumor necrosis factor-α (TNF-α) and paclitaxel on human adenoid cystic carcinoma cell line SACC-83 and its relationship with apoptosis. To study the effect of combination chemotherapy and biotherapy on adenoid cysts Cancer provides experimental evidence. Methods: The human adenoid cystic carcinoma cell line SACC-83 and control group primary cultured mouse lung fibroblasts were divided into 9 groups according to different drug concentration, the drug effect for 72 h. The inhibitory rate of cell proliferation was detected by 3H-TDR assay. The apoptosis rate and cell cycle were detected by flow cytometry. The morphological changes of apoptotic cells were observed by transmission electron microscope. Results: The inhibitory rate of cell proliferation in the combination group was significantly higher than that in the drug alone group (P <0.05). The inhibitory rate of cell proliferation in L929 cells was significantly lower than that in SACC-83 cells (P <0.05). Conclusion: TNF-α and paclitaxel have synergistic effects on the inhibition of SACC-83 cell proliferation and apoptosis induction.
