目的:研究4种黄荆子木脂类化合物(VBE-1,2,3,4)抑制人宫颈癌Hela细胞生长作用。方法:溴化标记尿嘧啶(BrdU)掺入法测定细胞核酸合成;平皿集落形成法和软琼脂培养集落形成法检测细胞生长。结果:4种黄荆子木脂类化合物对Hela细胞的核酸合成具有显著抑制作用,呈浓度依赖性;其中VBE-3的BrdU掺入抑制作用最强,IC50是0.93μg/mL。4种黄荆子木脂类化合物明显抑制Hela细胞的集落形成能力,VBE-3的效价强度最大,平皿集落形成法检测的IC50为0.13μg/mL;软琼脂培养集落形成法检测的IC50为0.57μg/mL。结论:黄荆子木脂类化合物具有抑制人宫颈癌Hela细胞生长作用。
OBJECTIVE: To study the inhibitory effect of four kinds of vitex lipid compounds (VBE-1, 2, 3, and 4) on the growth of human cervical cancer Hela cells. METHODS: The nucleic acid synthesis of cells was determined by bromine-labeled uracil (BrdU) incorporation method; plateau colony formation assay and soft agar culture colony formation assay were used to detect cell growth. RESULTS: Four kinds of oligosaccharides from Vitex negundo had a significant inhibitory effect on the nucleic acid synthesis of Hela cells in a concentration-dependent manner. Among them, BrdU incorporation of VBE-3 had the strongest inhibitory effect, IC50 was 0.93 μg/mL. The four jatropha lipid compounds significantly inhibited the colony forming ability of HeLa cells, and the potency of VBE-3 was the highest. The IC50 detected by plate colony forming assay was 0.13 μg/mL; the IC50 detected by soft agar cultured colony forming assay was 0.57 μg. /mL. Conclusion: J. vulgaris lignan compounds inhibit the growth of human cervical cancer Hela cells.
