目的 探讨错配修复基因 (MMR)蛋白MLH1在前列腺癌细胞株及前列腺癌标本的表达及其意义。方法 利用Western印迹法检测了前列腺癌细胞株中错配修复基因蛋白的丢失情况 ;用免疫组织化学法检测前列腺癌标本中错配修复蛋白的表达。结果 在所研究的 8个前列腺癌细胞株中均发现有错配修复基因的缺失 ;前列腺癌标本中也存在错配修复蛋白MLH1表达的减少或消失。结论 前列腺癌中存在错配修复基因的缺失 ,错配修复基因的缺失直接参与了前列腺癌的发生与发展。
Objective To investigate the expression and significance of mismatch repair gene (MMR) protein MLH1 in prostate cancer cell lines and prostate cancer specimens. Methods Western blotting was used to detect the loss of mismatch repair protein in prostate cancer cell lines. The expression of mismatch repair protein in prostate cancer specimens was detected by immunohistochemistry. Results The deletion of mismatch repair gene was found in all the 8 prostate cancer cell lines studied. There was also a decrease or disappearance of MLH1 expression in prostate cancer samples. Conclusion The deletion of mismatch repair gene exists in prostate cancer. The deletion of mismatch repair gene is directly involved in the occurrence and development of prostate cancer.
