目的探讨甜茶多酚对预防性高血脂大鼠的降血脂作用及其机制。方法采用水提法制备甜茶多酚浸膏,并过大孔树脂进行待试样品甜茶多酚的制备;取雄性SD大鼠60只,随机分为正常对照组、模型对照组、阳性对照组、甜茶多酚低、中、高剂量组(分别相当于生药量1.8、3.6、7.2 g·kg-1·d-1),正常对照组用普通饲料喂养,其他组均喂食高脂饲料。12周后测定大鼠血清中总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白(high-density lipoprotein,HDL-C)、低密度脂蛋白(low-density lipoprotein,LDL-C)、丙氨酸氨基转移酶(alanine aminotransferase,ALT)、天门冬氨酸氨基转移酶(asparate aminotransferase,AST)、脂肪酸合成酶(fatty acid synthetase,FAS)及肝脏中超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathion peroxidase,GSH-Px)含量及活性。结果甜茶多酚能显著降低模型组大鼠血清总胆固醇、甘油三酯、低密度脂蛋白、丙氨酸氨基转移酶、脂肪酸合成酶的含量,降低肝脏中丙二醛含量和谷胱甘肽过氧化物酶的活性。提高血清高密度脂蛋白含量和肝脏超氧化物歧化酶活性。结论甜茶多酚对高脂血症大鼠具有显著的降血脂效应。
Objective To investigate the hypolipidemic effect of sweet tea polyphenols on hyperlipidemic rats and its mechanism. Methods The water extract was used to prepare the sweet polyphenol extract and the macroporous resin was used to prepare the sweet polyphenols. Sixty male SD rats were randomly divided into normal control group, model control group and positive control group , Low, medium and high dose of sweet tea polyphenols (equivalent to 1.8,3.6,7.2 g · kg-1 · d-1, respectively). The normal control group was fed with normal diet, while the other groups were fed with high-fat diet. Serum levels of total cholesterol (TC), triglyceride (TG), high-density lipoprotein (HDL-C), low-density lipoprotein , LDL-C), alanine aminotransferase (ALT), asparate aminotransferase (AST), fatty acid synthetase (FAS) and superoxide dismutase superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-Px) and their activities. Results Sweet tea polyphenols could significantly reduce the content of serum total cholesterol, triglyceride, low density lipoprotein, alanine aminotransferase and fatty acid synthase, decrease the content of malondialdehyde and glutathione Oxidase activity. Increase serum high density lipoprotein and liver superoxide dismutase activity. Conclusion Sweet tea polyphenols have significant hypolipidemic effects on hyperlipidemic rats.
