目的:观察CLU基因干扰前后肾癌细胞株786-O的基因表达谱差异,探讨肾细胞癌与CLU基因相关基因及传导通路。方法:CLU基因干扰慢病毒载体及空病毒载体转染肾癌786-O细胞样本与RocheNimbleGen表达谱芯片杂交,比较干扰前后基因表达谱差异,并行GO分析及Pathway分析。结果:六份样本中,共筛选出差异表达基因1731个,其中共同上调基因1154个,共同下调基因577个。差异表达基因中生物进程、细胞成分、分子功能方面均以下调为主。通路分析显示29条通路上调,如细胞黏附、异体排斥、吞噬体。31条通路下调,如mTOR、MAPK、非小细胞肺癌。结论:采用全基因组芯片检测CLU基因干扰后肾癌786-O基因表达谱获得差异表达基因,为以CLU基因为靶点的肾癌基因研究提供了实验依据。
OBJECTIVE: To observe the gene expression profiles of 786-O cells before and after interference with CLU gene, and to explore the genes and pathways involved in renal cell carcinoma and CLU gene. Methods: CLU gene was transfected into leukemia cell line 786-O cells transfected with lentiviral vector and empty vector and then hybridized with Roche NimbleGen. The gene expression profiles were compared before and after interference. GO analysis and Pathway analysis were performed. Results: Among the six samples, 1731 differentially expressed genes were screened, of which 1154 genes were up-regulated and 577 genes were down-regulated. Differentially expressed genes in the biological process, cell composition, molecular function are mainly down. Pathway analysis showed that up to 29 pathways, such as cell adhesion, allograft rejection, phagosomes. 31 pathways down, such as mTOR, MAPK, non-small cell lung cancer. CONCLUSION: Genes differentially expressed in 786-O gene of renal cell carcinoma after CLU gene interference were detected by whole genome microarray. This study provides an experimental basis for the study of the gene of renal cell carcinoma with CLU gene as the target.
