目的:建立血中华法林浓度的毛细管胶束电动色谱分析方法,并对其进行评价。方法:用二氯甲烷提取血清中华法林,5-乙基-5-P-甲苯基巴比妥酸为内标,操作电压15kV,210nm波长检测。电泳缓冲液由25mmol/L硼酸盐,25mmol/L十二烷基硫酸钠,10mmol/L氯化钠(pH9.0)组成。结果:日内RSD1.4%~6.2%,日间RSD1.4%~4.7%,在0~10μg/ml范围内呈线性,最低血清华法林检测浓度为0.03μg/ml。测定结果与反相高效液相色谱法进行了比较,两种方法有很好相关性,r=0.9967(P>0.1)。结论:毛细管胶束电动色谱法精密度、准确度高,专一性强,是一种较理想的血药浓度检测方法。
OBJECTIVE: To establish capillary micellar electrokinetic chromatographic method for the determination of warfarin concentration in blood. Methods: Warfarin was extracted from the serum with dichloromethane, and 5-ethyl-5-p-tolylbarbituric acid was used as the internal standard. The operating voltage was detected at 15 kV and 210 nm. The electrophoresis buffer consists of 25 mmol / L borate, 25 mmol / L sodium lauryl sulfate, 10 mmol / L sodium chloride (pH 9.0). Results: Intraday RSD ranged from 1.4% to 6.2% and daytime RSD ranged from 0.4% to 4.7%. The linear range was from 0 to 10 μg / ml. The lowest serum warfarin concentration was 0.03 μg / ml. The results were compared with reversed-phase high-performance liquid chromatography, the two methods have a good correlation, r = 0.9967 (P> 0.1). Conclusion: Capillary micellar electrokinetic chromatography is more accurate, accurate and specific, and it is an ideal method for the determination of plasma concentration.
