目的研究含硒蛋白TRXR-1、TRXR-2、GPX1和GPX4在人体结肠癌组织不同病理阶段的表达水平。方法收集32名结肠癌患者行左半结肠切除术的新鲜组织(每个病期8名患者),作为对照的正常组织来自切除物的无癌部分。对同一样本使用ELISA,实时PCR和免疫组织化学(实时PCR和免疫组织化学技术仅用于T3和T4期肿瘤样本)3种方法检测。数据结果采用SPSS软件(10.0.7版本)和REST软件进行统计分析。结果结肠癌组织中的TRXR-1随着病情的恶化而升高,而正常组织与肿瘤组织的TRXR-2蛋白在任何时期的表达水平差异均不显著。与正常组织作比较,所有肿瘤组织的GPX1水平均升高;除T3期肿瘤组织外,其他病期肿瘤组织GPX4的表达水平比之正常组织显著上调。结论含硒蛋白在维持肿瘤细胞生存中起关键作用,可成为抗癌药物开发的潜在方向。
Objective To study the expression levels of selenoprotein TRXR-1, TRXR-2, GPX1 and GPX4 in human colon cancer tissues at different pathological stages. Methods Fresh tissues (8 patients per stage) were collected from 32 patients with colon cancer who underwent left hemocoeryctomy. Normal tissues served as controls from the non-cancerous portion of the excision. The same sample was detected by ELISA, real-time PCR and immunohistochemistry (real-time PCR and immunohistochemistry were used for T3 and T4 tumor samples only). Data Results SPSS software (version 10.0.7) and REST software for statistical analysis. Results The expression of TRXR-1 in colon cancer tissues increased with the progression of the disease, while the expression levels of TRXR-2 protein in normal and tumor tissues were not significantly different at any time. Compared with normal tissues, the GPX1 levels of all tumor tissues were increased. Except for T3 tumor tissues, the expression levels of GPX4 in tumor tissues at other stages were significantly up-regulated compared with those in normal tissues. Conclusion The selenoprotein plays a key role in maintaining the survival of tumor cells and may be the potential direction for anticancer drug development.
