目的观察基质金属蛋白酶(MMP)-2和金属蛋白酶抑制剂(TIMP)-1在类风湿关节炎(RA)滑膜组织及滑膜下组织的阳性表达,探讨其在RA发病中的作用。方法应用免疫组织化学ABC法检测65例RA组织MMP-2和TIMP-1表达情况,比较其在滑膜和滑膜下组织中的分布特点。另选取10例骨关节炎(OA)和7例半月板损伤的正常滑膜(NS)作对照。结果①MMP-2阳性表达率:RA 66%,OA 20%,NS 14%;TIMP-1阳性表达率:RA 63%,OA 30%,NS 0。②MMP-2在RA滑膜组织阳性表达率为40%,在滑膜下组织阳性表达率为61%;TIMP-1在RA滑膜组织的阳性表达率为60%,在滑膜下组织中的阳性表达率为35%。③MMP-2与TIMP-1之间,无明显的相关性表达。结论①MMP-2和TIMP-1在RA的发病机制中发挥作用;②RA滑膜的不同组织部位MMP和TIMP的表达存在差异。
Objective To investigate the expression of matrix metalloproteinase (MMP) -2 and metalloproteinase inhibitor (TIMP) -1 in synovial and synovial tissues of patients with rheumatoid arthritis (RA) and to explore its role in the pathogenesis of RA. Methods The expressions of MMP-2 and TIMP-1 in 65 cases of RA were detected by immunohistochemical ABC method. The distribution characteristics of MMP-2 and TIMP-1 in synovial tissue and synovial tissue were compared. Another 10 cases of osteoarthritis (OA) and 7 cases of normal meniscus injury synovial (NS) as a control. Results ①MMP-2 positive rate: RA 66%, OA 20%, NS 14%; TIMP-1 positive rate: RA 63%, OA 30%, NS 0. ② The positive expression rate of MMP-2 in RA synovial tissue was 40% and the positive expression rate in synovial tissue was 61%. The positive expression rate of TIMP-1 in RA synovial tissue was 60% The positive rate was 35%. ③MMP-2 and TIMP-1, no significant correlation between expression. Conclusions ①MMP-2 and TIMP-1 play a role in the pathogenesis of RA; ② The expression of MMP and TIMP in different tissues of RA synovial membrane are different.
