目的和方法 :采用电解Krebs灌流液产生自由基的方法 ,观察电解性自由基对豚鼠离体心脏冠脉内皮与气道上皮功能的影响。结果 :10mA直流电 ,持续 1- 2min电解灌流液产生的自由基导致离体心脏冠脉压升高约 4 4kPa ,同时使心肌收缩力下降 ,心肌组织过氧化脂质含量增高 ,而SOD活性降低 ;使离体支气管肺灌流压升高约 2 2 0kPa ,并对磷酸组胺反应性增高 3倍 ;电解灌流液的作用可被羟自由基清除剂甘露醇、DMSO抑制。结论 :电解性自由基可引起冠脉内皮与气道上皮损伤 ,使冠脉和气道反应性增高
OBJECTIVE: To observe the effect of free radical on the function of corneal endothelium and airway epithelium in isolated guinea pig hearts by using the method of free radical generation in Krebs perfusate. Results: Radicals generated by 10mA DC for 1 to 2min in electrolyzed perfusate increased the coronary pressure of isolated heart by about 4 4 kPa, decreased the contractility of myocardium and the content of peroxidation lipid and the activity of SOD in myocardium. In vitro bronchial hyperpermeability perfusion pressure increased about 220kPa, and 3-fold increased histamine reactivity; electrolytic perfusate by hydroxyl radical scavengers mannitol, DMSO inhibition. Conclusion: Electrolytic free radicals can cause injury of coronary artery endothelium and airway epithelium, which leads to increased coronary and airway responsiveness
