目的:探讨不同抗褐化剂对明党参悬浮细胞数目、发生褐化反应底物酚类及明党参的有效成分香豆素的影响。方法:在轻度褐化的明党参悬浮细胞中添加不同浓度的维生素C(VC)、活性炭(AC)、酸水解酪蛋白(AHC)、硫代硫酸钠(Na_2S_2O_3)以及聚乙烯吡咯烷酮(PVP),利用紫外分光光度法和高效液相色谱法检测酚类、总香豆素及佛手酚、佛手柑内酯含量。结果:低浓度PVP、VC均不同程度促进明党参悬浮细胞的生长;不同抗褐化剂对酚类含量高低影响不同;VC处理后悬浮细胞中总香豆素含量明显增高,其中用2 mg/m L的VC处理后,总香豆素增长率为51.53%,是对照组的4.8倍,将该浓度VC添加到以水杨酸作为诱导子的明党参悬浮培养液中,改善了水杨酸的促进褐化现象,同时佛手酚、佛手柑内酯的含量提高了25.96%、33.33%。结论:VC为本研究中的最佳抗褐化剂,能在抑制悬浮细胞褐化的同时,有效地促进细胞生长和次生代谢产物的积累。
Objective: To investigate the effects of different anti-browning agents on the number of suspended cells in Cynanchi paniculatum, the phenolic compounds of coumarin and the active ingredient coumarin in Codonopsis. The content of total coumarins in suspension cells increased significantly after treatment with VC, m VC, the total coumarin growth rate was 51.53%, which is 4.8 times that of the control group.
