目的:建立HPLC同时测定山蜡梅叶中4种有效成分的含量。方法:采用依利特hypersil ODS2(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-水,梯度洗脱,流速1.0 mL.min-1,检测波长344 nm,柱温40℃。结果:东莨菪内酯在5.665~56.65 mg.L-1线性关系良好(r=0.999 9),平均回收率为97.06%,RSD 1.62%(n=6),异嗪皮啶在2.431 2~24.312 mg.L-1线性关系良好(r=0.999 9),平均回收率为102.86%,RSD 0.93%(n=6),滨蒿内酯在2.444~24.44 mg.L-1线性关系良好(r=0.999 9),平均回收率为102.18%,RSD 1.81%(n=6),芦丁在9.012~90.12 mg.L-1线性关系良好(r=0.999 2),平均回收率为104.44%,RSD 4.2%(n=6)。结论:方法操作简便,结果准确,重复性好,可用来控制山蜡梅药材的质量。
OBJECTIVE: To establish an HPLC method for the simultaneous determination of four active ingredients in the leaves of Pseudostellariae sinensis. Methods: The elution was performed on a hypersil ODS2 column (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of acetonitrile and water with a gradient of 1.0 mL · min-1. The detection wavelength was set at 344 nm and the column temperature was set at 40 ℃. Results: Scopolactone showed good linearity (r = 0.999 9) between 5.665 and 56.65 mg · L -1, with an average recovery of 97.06% and a RSD of 1.62% (n = 6) The linear relationship was good (r = 0.999 9), the average recovery was 102.86%, RSD 0.93% (n = 6), scopolamine was good at 2.444 ~ 24.44 mg.L-1 (r = 0.999 9). The average recovery was 102.18%, RSD 1.81% (n = 6) and rutin was 9.012 ~ 90.12 mg.L-1. The average recovery was 104.44%, RSD 4.2 % (n = 6). Conclusion: The method is simple, accurate, reproducible and can be used to control the quality of medicinal materials.
