采用静置贴壁培养法,体外长期培养了胎儿、儿童和成人骨髓基质细胞,并将传至5代以上的肌样骨髓基质细胞采用微量细胞病变(CPE)法,对单纯疱疹病毒进行了敏感性试验。结果:人骨髓基质肌样细胞对Ⅰ型和Ⅱ型单纯疱疹病毒均敏感,能产生明显的CPE,其效价(TCID50)可达10-3~10-4,其中胎儿骨髓基质肌样细胞对病毒敏感性比儿童高2倍,比成人高4倍。提示此法可为人骨髓细胞体外长期培养提供简便方法,且为该细胞对病毒的敏感性提供实验依据,并为该细胞的利用开辟了新的应用前景。
Adopting static adherent culture method, fetal, children and adult bone marrow stromal cells were cultured in vitro for a long time, and myogenic bone marrow stromal cells which were passaged to more than 5th generation were sensitized to herpes simplex virus Sexual test. Results: Human bone marrow stromal myoblast cells were sensitive to both herpes simplex virus type I and type II, and could produce significant CPE. The titer (TCID50) was 10-3 ~ 10-4. Virus sensitivity 2 times higher than children, 4 times higher than adults. It is suggested that this method can provide a simple method for long-term culture of human bone marrow cells in vitro and provide an experimental basis for the sensitivity of the cells to the virus and open up new application prospects for the utilization of the cells.
