目的探讨了Fas基因是否参与重组人粒细胞集落刺激因子(rhGCSF)对白血病细胞的诱导凋亡过程。方法利用脂质体(DOTAP)介导的基因转染技术将Fas基因转入HL60细胞中,并经原位杂交、Westernblot及流式细胞术(FCM)证实已成功建立了Fas高表达的HL60细胞株,用FCM对Fas基因在rhGCSF诱导白血病细胞凋亡过程中的作用进行了探讨。结果相同浓度的rhGCSF作用相同时间后转染的HL60细胞凋亡率较未转染的HL60细胞凋亡率明显增加,且随rhGCSF作用时间延长,Fas在HL60细胞中的表达逐渐增加。结论Fas基因是凋亡诱导基因,rhGCSF诱导白血病细胞凋亡依赖Fas径路传递凋亡信号。
Objective To investigate whether Fas gene is involved in the induction of apoptosis of leukemia cells by recombinant human granulocyte colony-stimulating factor (rhG-CSF). Methods Fas gene was transfected into HL60 cells by lipofectamine (DOTAP)-mediated gene transfection. The expression of Fas was successfully established by in situ hybridization, Western blot, and flow cytometry (FCM). The role of Fas gene in the apoptosis of leukemic cells induced by rhGCSF was investigated in HL60 cell line. Results The apoptosis rate of HL-60 cells transfected with the same concentration of rhG-CSF was significantly higher than that of non-transfected HL-60 cells at the same time. With the prolongation of rhG-CSF action time, Fas in HL-60 The expression in cells gradually increases. Conclusion Fas gene is an apoptosis-inducing gene. The apoptosis of leukemic cells induced by rhGCSF is dependent on Fas pathway to transmit apoptotic signals.
