目的:观察中成药香砂六君子丸对脾虚痰浊证大鼠心肌内皮素受体(ETAR)及血管紧张素受体(AT1R)表达的影响,以期探究其治疗脾虚痰浊证的作用机制。方法:将18只SD大鼠,随机分为空白对照组、脾虚痰浊组和治疗组(香砂六君子丸)。每组6只,制备大鼠脾虚痰浊证模型,采用ELISA法检测大鼠血清中ET、AngⅡ水平,RT-PCR和Western Blot检测心肌组织中ETAR和AT1R的mRNA及蛋白表达水平。结果:ELISA结果提示,与空白对照组相比,脾虚痰浊组大鼠血清中ET、AngⅡ含量显著升高;与脾虚痰浊组相比,治疗组大鼠血清中ET、AngⅡ含量显著降低。PCR结果提示,与空白对照组相比,脾虚痰浊组心肌组织中ETARmRNA和AT1RmRNA含量显著减少;与脾虚痰浊组相比,治疗组心肌组织中ETARmRNA和AT1RmRNA含量显著升高。Western Blot结果提示,与空白对照组相比,脾虚痰浊组心肌组织中ETAR和AT1R蛋白表达量显著减少;与脾虚痰浊组相比,治疗组心肌组织中ETAR和AT1R蛋白表达量显著升高。结论:香砂六君子丸能够改善脾虚痰浊证,其机制可能与调节大鼠心肌ETAR和AT1R表达有关。
OBJECTIVE: To observe the effect of Xiangsha Liujunzi Pill, a Chinese patent medicine, on the expression of myocardial endothelin receptor (ETAR) and angiotensin receptor (AT1R) in rats with spleen deficiency and turbidity in order to explore its mechanism of action in treating spleen deficiency and turbidity syndrome. Methods: Eighteen SD rats were randomly divided into blank control group, spleen deficiency turbid group and treatment group (Xiangsha Liujunzi Pills). Six rats in each group were used to prepare a rat model of spleen deficiency and stasis syndrome. ET and Ang II levels in serum of rats were detected by ELISA. The mRNA and protein expressions of ETAR and AT1R in myocardial tissue were detected by RT-PCR and Western Blot. Results: The results of ELISA suggested that compared with the control group, the contents of ET and Ang II in the serum of the spleen-deficient and cloudy group increased significantly. Compared with the spleen deficiency and phlegm group, the contents of ET and Ang II in the serum of the treatment group were significantly lower. The results of PCR indicated that compared with the control group, the contents of ETAR mRNA and AT1R mRNA in the myocardial tissue of spleen deficiency and phlegm group were significantly reduced. Compared with spleen deficiency and phlegm group, the contents of ETAR mRNA and AT1R mRNA in the myocardial tissue of the treatment group were significantly increased. Western Blot results indicated that compared with the blank control group, the expression of ETAR and AT1R protein in the myocardial tissue of spleen deficiency and turbidity group was significantly reduced; compared with the spleen deficiency and phlegm group, the expression levels of ETAR and AT1R protein in the myocardial tissue of the treatment group were significantly increased. . Conclusion: Xiangsha Liujunzi Pill can improve the syndrome of spleen deficiency and phlegm, and its mechanism may be related to the regulation of myocardial ETR and AT1R expression.
