目的:测定人鼻咽癌细胞CNE1、CNE2、Hone-1及C666-1的放射生物学特征参数。方法:采用平板集落形成法测定CNE1、CNE2、Hone-1及C666-14种鼻咽癌细胞在0、1、2、3、4、6、8、10Gy照射后的存活分数;通过单击-多靶模型和线性-二次模型分别拟合细胞存活曲线;并计算D0、Dq、N、α、β、α/β值及2Gy照射时的存活分数(SF2)等放射生物学参数值。结果:4种细胞在2种数学模型拟合的细胞存活曲线差异均有统计学意义(P均<0.05);其参数D0、Dq、N、α、β、α/β和SF2分别在1.818~2.379、0.192~0.880、1.276~2.388、0.085~0.392、0.026~0.036、2.36~15.08和0.416~0.733。结论:较大的放射敏感性差异可能存在于不同甚至相同分化程度的鼻咽癌细胞之间,提示临床鼻咽癌放疗剂量的个体化需求。
Objective: To determine the radiobiological parameters of human nasopharyngeal carcinoma cells CNE1, CNE2, Hone-1 and C666-1. Methods: Survival scores of CNE1, CNE2, Hone-1 and C666-14 nasopharyngeal carcinoma cells after 0, 1, 2, 3, 4, 6, 8, 10 Gy irradiation were determined by plate- Multi-target model and linear-quadratic model were used to fit the cell survival curve respectively. The radioactivity parameters such as D0, Dq, N, α, β, α / β and surviving fraction (SF2) at 2Gy irradiation were calculated. Results: The cell survival curves of four kinds of cells in two kinds of mathematical models were all statistically significant (P <0.05). The parameters D0, Dq, N, α, β, α / β and SF2 were between 1.818 ~ 1.92 to 0.880, 1.276 to 2.388, 0.085 to 0.392, 0.026 to 0.036, 2.36 to 15.08, and 0.416 to 0.733. Conclusion: The larger difference in radiosensitivity may exist between different or even the same degree of differentiation of nasopharyngeal carcinoma cells, suggesting that the dose of radiotherapy for individual NPC needs.
