目的:探讨血管内皮生长因子D(VEGF-D)表达与胃癌淋巴结转移、淋巴管生成的关系。方法:用组织芯片及免疫组化技术检测VEGF-D在胃癌组织及正常组织中的表达,以淋巴管特异性标志D2-40显示并计数淋巴管密度(LVD)。结果:VEGF-D在淋巴结转移组中呈高表达(68.05%,49/72),显著高于无淋巴结转移组(29.03%,9/31),χ2=13.41,P=0.001。VEGF-D表达与淋巴结转移(χ2=13.41,P=0.001)、TNM分期(χ2=22.05,P=0.000)有关,与年龄(χ2=2.68,P=0.101)、性别(χ2=0.42,P=0.518)以及浸润深度(χ2=0.13,P=0.718)无关。VEGF-D阳性组LVD值(35.42±10.33)较阴性组(17.71±5.28)显著增高,t=4.71,P=0.000。结论:VEGF-D高表达与胃癌淋巴结转移密切相关,选择性阻断VEGF-D的作用对控制胃癌淋巴结转移可能是一个有效的途径。
Objective: To investigate the relationship between the expression of vascular endothelial growth factor D (VEGF-D) and lymph node metastasis and lymphangiogenesis in gastric cancer. Methods: The expression of VEGF-D in gastric cancer tissues and normal tissues was detected by tissue microarray and immunohistochemistry. The lymphatic vessel density (LVD) was detected and counted by lymphatic-specific marker D2-40. Results: VEGF-D was highly expressed in lymph node metastasis group (68.05%, 49/72), which was significantly higher than that in non-lymph node metastasis group (29.03%, 9/31), χ2 = 13.41, P = 0.001. The expression of VEGF-D was correlated with lymph node metastasis (χ2 = 13.41, P = 0.001) and TNM stage (χ2 = 22.05, P = 0.000) 0.518) and depth of invasion (χ2 = 0.13, P = 0.718). The LVD of VEGF-D positive group (35.42 ± 10.33) was significantly higher than that of negative group (17.71 ± 5.28), t = 4.71, P = 0.000. CONCLUSION: High expression of VEGF-D is closely related to lymph node metastasis of gastric cancer. Selective blocking of VEGF-D may be an effective way to control lymph node metastasis of gastric cancer.
