将小叶杨×美杨、小叶杨×胡杨、小叶杨×大叶杨、小叶杨×椅杨、箭杆杨×大叶杨和741×[银白杨×(山杨×小叶杨)]的杂种胚珠进行离体培养,全部成功地获得了杂种植株。以1/2MS为基本培养基,进行了下列试验:1.不同浓度IAA和BA对诱导不定芽和绿苗发生的效应,结果最适分化培养基为1/2MS+IAA0.01mg/1+BA0.1mg/1;除去IAA,仅附加BA0.1mg/1,也是有效的;2.不同浓度NAA和IBA对生根的影响,试验证明最适生根培养基为1/2MS+NAA0.01—0.02mg/1或1/2MS+IBA0.1—0.5mg/1。接种时杂种胚珠内胚的发育阶段对萌发率及以后的生长有显著影响:含早期心形胚的杂种胚珠,萌发率只有33%,小苗弱而且很难长大;含中期或后期心形胚、鱼雷胚及子叶伸长期幼胚的杂种胚珠,萌发率在50%以上,最高可达100%,小苗健壮,容易分化芽和绿苗丛。胚根端和下胚轴的横切面显示:表皮、皮层、内皮层和中柱鞘的细胞脱分化成为分生细胞。这些分生细胞分裂活动的结果,形成了不定芽。

Cross-sections of the radicle and hypocotyls show that cells of the epidermis, cortex, endothelium, and pericycle are dedifferentiated into meristematic cells. As a result of these meristematic cell divisions, adventitious buds are formed.
