目的:研究秦皮总香豆素对急性痛风性关节炎模型大鼠血清IL-1β、TNF-α的影响,探讨秦皮总香豆素其治疗急性痛风性关节炎的作用机理。方法:将雄性Wistar大鼠90只随机分为6组:正常对照组、模型组、秋水仙碱组、秦皮高剂量、秦皮中剂量、秦皮小剂量组。采用先给干预性药物4d,然后尿酸钠关节内注射造模法致痛风性关节炎模型,72h后取大鼠血清,应用ELISA法测定IL-1β、TNF-α的含量。结果:模型组IL-1β和TNF-α的水平明显高于正常组;秦皮总香豆素能够显著抑制急性痛风性关节炎大鼠血清IL-1β和TNF-α的表达,与秋水仙碱作用相近。结论:秦皮总香豆素对急性痛风性关节炎具有良好的治疗作用,其作用机制与抑制血清IL-1β、TNF-α的产生有关。
OBJECTIVE: To study the effect of total coumarins of Qinhuang Pear on serum IL-1β and TNF-α in rats with acute gouty arthritis and to explore the mechanism of total coumarin in treatment of acute gouty arthritis. Methods: Ninety male Wistar rats were randomly divided into 6 groups: normal control group, model group, colchicine group, high dosage of Qinpi, medium dosage of Qinpi and low dosage of Qinpi. Gouty arthritis model was induced by intraperitoneal injection of sodium urate for 4 days, and serum of rats was taken after 72 hours. The levels of IL-1β and TNF-α were determined by ELISA. Results: The levels of IL-1β and TNF-α in the model group were significantly higher than those in the normal group. Total coumarin in Qinpi skin group could significantly inhibit the expression of IL-1β and TNF-α in serum in rats with acute gouty arthritis, similar. Conclusion: The total coumarin of Qinpi has a good therapeutic effect on acute gouty arthritis, and its mechanism of action is related to inhibiting the production of IL-1β and TNF-α.
