采用薄层扫描法测定小蔓长春花组织培养物中长春胺的含量。应用高效硅胶G薄层板,展开剂为石油醚-氯仿-甲醇(10:10:1.2)。在λs=490nm,λR=550nm波长处扫描。长春胺标准品的线性范围为0.535~3.745μg;平均回收率为98.68%,RSD=1.9%(n=6)。小蔓长春花组织培养物经碱性氯仿和2%硫酸提取后点样,按随行外标两点法进行测定,计算样品的含量。测得在固体和液体培养基B5a上生长的由根、茎、叶诱导产生的培养物中长春胺含量分别为:0.62±0.22‰、0.31±0.07‰、0.39±0.10‰和0.59±0.09‰、0.56±0.15‰、0.52±0.07‰。
The content of vincamine in the tissue culture of Vinca minor was measured by TLC scanning. A high-efficiency silica gel G thin plate was used and the developing solvent was petroleum ether-chloroform-methanol (10:10:1.2). Scanned at a wavelength of λs=490 nm, λR=550 nm. The linear range of the vincamine standard was 0.535 to 3.745 μg; the average recovery was 98.68%, and the RSD was 1.9% (n=6). The tissue culture of Vinca minors was spotted after extraction with basic chloroform and 2% sulfuric acid. The content was determined according to the two-point method accompanied by external standards. The contents of vincamine in cultures induced from roots, stems, and leaves grown on solid and liquid media B5a were found to be 0.62±0.22 ‰, 0.31±0.07 ‰, 0, respectively. 39±0.10 ‰ and 0.59±0.09 ‰, 0.56±0.15 ‰, and 0.52±0.07 ‰.
