目的建立虎柏烧伤酊中大黄素、虎杖苷和盐酸小檗碱定量测定方法。方法采用Eclipse XDB-C18(150 mm×4.6mm,5μm)色谱柱,流动相为乙腈-0.1%磷酸水溶液,梯度洗脱,体积流量1.0 mL/min,检测波长265 nm(盐酸小檗碱)、306 nm(大黄素和虎杖苷),柱温30℃。结果大黄素在6.3~200.0 ng、虎杖苷在9.6~308.0 ng、盐酸小檗碱在5.0~160.0ng有良好的线性关系,样品中大黄素、虎杖苷和盐酸小檗碱的平均加样回收率分别为99.91%、99.51%和99.16%,RSD分别为1.12%、0.84%和1.17%。结论本法结果准确,精密度及重复性较好,可作为虎柏烧伤酊的质量控制方法。
Objective To establish a method for the quantitative determination of emodin, polydatin and berberine hydrochloride in Burdock tincture. Methods The mobile phase was acetonitrile-0.1% phosphoric acid aqueous solution with gradient elution at a flow rate of 1.0 mL / min using Eclipse XDB-C18 column (150 mm × 4.6 mm, 5 μm) with a detection wavelength of 265 nm (berberine hydrochloride) 306 nm (emodin and polydatin), column temperature 30 ℃. Results Emodin had a good linear relationship between 6.3 and 200.0 ng of polydatin, between 9.6 and 308.0 ng of polydatin and between 5.0 and 160.0 ng of berberine hydrochloride. The average recoveries of emodin, polydatin and berberine were 99.91%, 99.51% and 99.16% respectively, with RSDs of 1.12%, 0.84% and 1.17% respectively. Conclusion This method is accurate, accurate and reproducible. It can be used as quality control method for Burkind’s tincture.
