目的 观察不同浓度的降钙素基因相关肽 ( CGRP)对缺氧状态下人脐静脉血管内皮细胞的影响 .方法 用培养的人脐静脉血管内皮细胞建立缺氧模型 ,观察血管内皮细胞在缺氧状态下细胞膜流动性 ,5 1铬释放率 ,台芬蓝摄取率和细胞内钙 ,镁含量的变化以及 1× 10 - 9,1× 10 - 8,1× 10 - 7mol.L- 1 CGRP对血管内皮细胞的影响 .结果 1× 10 - 9,1×10 - 8,1× 10 - 7mol.L- 1 CGRP可降低缺氧 12 0 min时细胞膜流动性 ,5 1 铬释放率 ,台芬蓝摄取率及减少细胞内镁的丢失 ,1× 10 - 8mol.L- 1 CGRP的效果最好 .结论 CGRP对血管内皮细胞缺氧损伤具有直接保护作用 ,在一定范围内存在浓度效应关系
Objective To observe the effects of different concentrations of calcitonin gene related peptide (CGRP) on human umbilical vein endothelial cells (HUVECs) under hypoxia condition.Methods Cultured human umbilical vein endothelial cells (HUVECs) were used to establish hypoxia model to observe the effects of CGRP on hypoxia (1 × 10 - 9, 1 × 10 - 8, 1 × 10 - 7mol.L - 1 CGRP) on the membrane fluidity, the release rate of 5 1 chromium, the uptake of tenpirin and the intracellular calcium and magnesium contents Vascular endothelial cells.Results 1 × 10 - 9, 1 × 10 - 8, 1 × 10 - 7mol.L - 1 CGRP could reduce the cell membrane fluidity, release rate of chromium 5 1, Uptake rate and the loss of intracellular magnesium, 1 × 10 - 8mol.L-1 CGRP was the best.Conclusion CGRP has a direct protective effect on vascular endothelial cell injury induced by hypoxia, and there is a concentration-effect relationship in a certain range
