目的初步探讨蛋白质组技术在血清研究中的应用,建立低分化胃癌患者血清和健康者血清双向凝胶电泳图谱;筛查并鉴定低分化胃癌患者血清和健康者血清中的差异表达蛋白质,期望从中发现有意义的胃癌分子标志物。方法采用双向凝胶电泳(2-DE)分离低分化胃癌患者血清和健康者血清总蛋白,银染显色,PDQuest软件分析,从中选取差异表达蛋白质,通过基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)鉴定差异表达的蛋白质。结果获得重复性和分辨率较好的血清双向凝胶电泳图谱;鉴定得到6个仅在胃癌组血清中表达的蛋白质和4个仅在对照组血清中表达的蛋白质。结论在低分化胃癌患者血清与健康者血清中存在差异表达蛋白质,利用蛋白质组学这一高通量的筛查技术可望从中发现与低分化胃癌相关的标志物。
Objective To explore the application of proteome technology in serum research, establish two-dimensional gel electrophoresis profiles of serum and healthy healthy people in poorly differentiated gastric cancer, and screen and identify differentially expressed proteins in serum of healthy and poorly differentiated gastric cancer patients. Discovered meaningful gastric cancer molecular markers. Methods 2-DE was used to separate the serum total protein from healthy and poorly differentiated gastric cancer patients. Silver staining was used for color development. PDQuest software was used to analyze the differentially expressed proteins. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI) was used. -TOF-MS) identify differentially expressed proteins. Results Two-dimensional gel electrophoresis profiles with reproducibility and resolution were obtained. Six proteins expressed only in the serum of gastric cancer group and four proteins expressed only in the serum of the control group were identified. Conclusion There are differentially expressed proteins in the serum of poorly differentiated gastric cancer patients and healthy individuals. Proteomics, a high-throughput screening technique, is expected to find markers associated with poorly differentiated gastric cancer.
