目的 研究莪术油注射液对大鼠球囊损伤血管新生内膜形成及血管重构的作用以及Ⅰ、Ⅲ型胶原的影响。方法 用球囊导管损伤大鼠左颈总动脉制成再狭窄模型 ,术后 14d用计算机图像分析系统分析形态学及组织学的变化 ,苦味酸天狼猩红偏振光显示法结合图像分析检测各组Ⅰ型及Ⅲ型胶原的变化。结果 术后第 14天 ,偏光显微镜下 ,模型组Ⅰ、Ⅲ型胶原较假手术组明显增多、增粗、变亮、紊乱 ,其分布以外膜为主 ,莪术油组与模型组相比 ,Ⅰ、Ⅲ型胶原明显增少、变细、变暗、排列较整齐 ,其分布也以外膜为主 ;模型组较假手术组腔面积、外弹力板周长、内弹力板周长明显减少 ,Ⅰ、Ⅲ型胶原的面积明显增加 ;莪术油组和模型组比较 ,最大内膜厚度、新生内膜面积及Ⅰ、Ⅲ型胶原面积明显减少 ,腔面积、外弹力板周长、内弹力板周长明显增加 ,与假手术组比较 ,差异无显著性 ;3组胶原Ⅰ /Ⅲ差异无显著性。结论 Ⅰ、Ⅲ型胶原排列的紊乱 ,含量的增加是再狭窄形成的重要机制 ,其比值可能并不重要。莪术油注射液显著抑制了球囊损伤大鼠颈动脉内膜增生和血管重构 ,作用机制可能与抑制Ⅰ、Ⅲ型胶原积聚有关。
Objective To study the effect of Zedoary oil injection on the neointimal formation and vascular remodeling of rat balloon injury and the effects of type I and type III collagen. Methods The left common carotid artery was injured by a balloon catheter to make a restenosis model. Morphological and histological changes were analyzed by a computerized image analysis system 14 days after surgery. Picrosirius red polarized light display combined with image analysis was used to detect each group. Type I and Type III collagen changes. RESULTS: On the 14th day after the operation, under the polarizing microscope, collagen I and III in the model group were significantly increased, thickened, brightened, and disordered compared with the sham operation group. The distribution was mainly in the outer membrane, and compared with the model group in the zedoary oil group, I Collagen type III and collagen III were significantly smaller, thinner, darker, and arranged more neatly, and their distribution was also mainly in the outer membrane; the luminal area of the sham-operated group, the perimeter of the outer elastic plate, and the perimeter of the inner elastic plate were significantly reduced in the model group compared with the sham group. The area of Type III and Type III collagen increased significantly; compared with the model group, the maximum thickness of intima, area of neointimal and area of type I and III collagen were significantly reduced, and the area of the cavity, the length of the external elastic plate, and the length of the internal elastic plate were significantly reduced. Significantly increased, compared with the sham group, the difference was not significant; there was no significant difference in collagen I/III between the 3 groups. Conclusion The disorder of arrangement of type I and type III collagen and the increase of its content are important mechanisms for the formation of restenosis. The ratio may not be important. Zedoary oil injection significantly inhibited carotid artery intimal hyperplasia and vascular remodeling in rats with balloon injury. The mechanism may be related to the inhibition of type I and type III collagen accumulation.
