目的:探讨鲜地黄、生地黄和熟地黄对大鼠肝组织皮质酮降解的影响。方法:用1640培养液制备肝组织悬液,其中添加不同浓度的皮质酮,以及鲜地黄、生地黄和熟地黄提取液,培养45 min后,用醋酸乙酯提取,以皮质醇作为内标,高效液相色谱法测定皮质酮的含量。结果:当皮质酮质量浓度为0.1,0.5 mg.L-1时,其降解率分别为71.1%,42.9%,添加不同剂量鲜地黄、生地黄或熟地黄后,皮质酮降解率与对照比较无显著差异。结论:地黄不同炮制品对大鼠肝脏皮质酮降解无影响,抗氧化可能是其抗炎机制之一。
Objective: To investigate the effects of fresh rehmanniae, rehmannia root and rehmannia root on the degradation of corticosterone in rat liver tissue. Methods: The suspension of liver tissue was prepared with 1640 culture medium, and different concentrations of corticosterone and fresh extract of Rehmannia glutinosa, Rehmannia glutinosa and Rehmannia glutinosa were added. After cultured for 45 min, the mixture was extracted with ethyl acetate and cortisol was used as internal standard. Determination of Corticosterone by High Performance Liquid Chromatography. Results: The degradation rates of corticosterone were 71.1% and 42.9% when the corticosterone concentrations were 0.1 and 0.5 mg.L-1 respectively. Compared with the control, the degradation rates of corticosterone were not significantly different after adding different dosages of fresh yellow, rhubarb or rehmannia root Significant differences. Conclusion: Different preparations of Rehmannia glutinosa have no effect on hepatic corticosterone degradation in rats, and anti-oxidation may be one of its anti-inflammatory mechanisms.
