背景:复方黄连能够抑制鼻咽癌细胞移植瘤的生长,但其作用的机制并不很清楚。目的:探讨复方黄连抑制鼻咽癌细胞移植瘤生长的机制。设计:随机对照的实验研究。地点、材料和方法:实验在中南大学湘雅医学院动物实验中心完成。实验选用10只Balb/C裸鼠,鼠龄3~4周。饲养1周后,在每只裸鼠的前腋下接种0.5mL(0.5×107个)HNE1细胞。移植瘤形成后,根据移植瘤的大小,按数字法将裸鼠随机分成实验组和对照组,每组5只。实验组灌喂复方黄连,对照组裸鼠仅灌喂无菌水。从瘤组织提取RNA,经返转录荧光标记作为探针与cDNA芯片杂交后用激光共聚焦扫描仪分析基因表达并通过返转录-聚合酶链式反应技术进一步证实基因的表达。主要观察指标:复方黄连处理后HNE1鼻咽癌细胞移植瘤的基因表达。结果:复方黄连处理30d后,移植瘤明显减小,同时有335条基因的表达发生改变,即242条基因表达下调和93条基因表达上调。通过RT-PCR分析,6条基因证实为真正差异表达。其中,MAD3,H731和EGR-α基因mRNA表达增高,而TRAF5,P68激酶和CHK1基因mRNA的表达下降。结论:复方黄连能够抑制HNE1鼻咽癌裸鼠移植瘤的生长,对HNE1鼻咽癌裸鼠移植瘤的抑制作用可能与基因表达的改变有关。
BACKGROUND: Coptis can inhibit the growth of nasopharyngeal carcinoma xenografts, but its mechanism is not clear. Objective: To investigate the mechanism of compound Coptidis Inhibit the growth of nasopharyngeal carcinoma xenografts. Design: Randomized controlled experimental study. Location, Materials and Methods: The experiment was performed at the Animal Experiment Center of Xiangya Medical College of Central South University. Ten Balb / C nude mice were selected for experiment, and were 3 to 4 weeks old. After one week of feeding, 0.5 mL (0.5 x 107) HNE1 cells were inoculated into the anterior armpit of each nude mouse. After the formation of xenografts, nude mice were randomly divided into experimental group and control group according to the size of the transplanted tumor, with 5 in each group. Experimental group fed Coptis, nude mice in the control group were fed sterile water. RNA was extracted from the tumor tissue, and the gene expression was analyzed by laser confocal scanner after hybridization with cDNA chip by reverse transcribed fluorescent label and the gene expression was further confirmed by reverse transcription-polymerase chain reaction. MAIN OUTCOME MEASURES: Gene expression of HNE1 nasopharyngeal carcinoma xenografts after compound Coptis chinensis treatment. Results: After treated with Coptis chinensis for 30 days, the number of xenografts decreased significantly, while the expression of 335 genes changed. That is, 242 genes were down-regulated and 93 genes were up-regulated. By RT-PCR analysis, the six genes were confirmed as truly differentially expressed. Among them, MAD3, H731 and EGR-α gene mRNA expression increased, while TRAF5, P68 kinase and CHK1 gene mRNA expression decreased. CONCLUSION: Fufang Huanglian can inhibit the growth of HNE1 nasopharyngeal carcinoma xenografts in nude mice, which may be related to the change of gene expression in HNE1 nasopharyngeal carcinoma xenografts in nude mice.
