目的:观察RT-PCR检测沙参麦冬汤对胃癌小鼠模型癌基因K-ras的影响。方法:建立MFC胃癌小鼠模型,予沙参麦冬汤灌胃治疗4周后,采用荧光定量RT-PCR检测K-ras基因的表达水平。结果:干预组经沙参麦冬汤处理4周后,K-ras mRNA的表达显著低于未用沙参麦冬汤处理的模型组(t=2.724.,P=0.018)。结论:沙参麦冬汤可以下调MFC胃癌小鼠K-ras基因的表达,其机制可能为抑制瘤体组织中K-ras基因的过表达,干预K-ras的突变或稳定Kras基因的表达来调控癌变的进展。
OBJECTIVE: To observe the effect of Radix Scutellariae Radix Ophiopogonis decoction on the oncogene K-ras in gastric cancer mouse model by RT-PCR. Methods: The murine model of gastric cancer was established. The rats were treated with Radix Stemi Radix Ophiopogonis for 4 weeks, and the expression of K-ras gene was detected by real-time RT-PCR. Results: The expression of K-ras mRNA in the intervention group was significantly lower than that in the untreated group (t = 2.724, P = 0.018) after 4 weeks of treatment. Conclusion: Ssangyin Maodong Decoction can down-regulate the expression of K-ras gene in MFC gastric cancer mice by down-regulating the expression of K-ras gene or K-ras gene in the tumor tissue Regulate the progress of cancer.
