为了排除柑桔多胚性对杂交育种的干扰,提高育种效率,进行了成熟种子胚的分离培养研究。证明:一、二级大胚采用普通砂培法,成苗率可达90%以上;在含有GA_3(1ppm)的MS培养基上,三、四级小胚成苗率为80%以上,五、六级小胚也有52.1%发育成苗,为提高杂种实生苗的获得率提供了有效途径。 进一步探讨了各种培养基添加物的生理效应,提出以IAA(1ppm)+CH(400ppm)替代GA_3(1ppm)具有更好的效果,培养的小苗生育健壮,根系发达,为入土成活打下了良好的基础。
In order to eliminate the interference of polyembryony on hybrid breeding and improve the breeding efficiency, the research on the isolation and culture of mature seed embryo was carried out. The results showed that the first and second large embryos were cultured by ordinary sand culture method, the rate of seedling formation was over 90%. On the MS medium containing GA_3 (1ppm), the germination rate of three or four small embryos was over 80% , And six small embryos also developed 52.1%, which provided an effective way to improve the yield of hybrid seedlings.
