利用ISSR分子标记技术探讨了天女花与广玉兰杂交后代9株幼苗的遗传多样性。9株F_1代幼苗根据生长健壮的情况依次标记为SA、SB、SC、SD、SE、SF,、SG、SH、SI。选用10个ISSR引物对亲本以及9株F_1代幼苗的基因组进行分析。结果表明,11株植物共扩增出96条DNA条带,其中多态性条带为85条,占总条带的88.5%。UPGMA聚类结果表明所有F_1代幼苗与六倍体父本更为相似,与二倍体的母本遗传距离较远。其中SC无论从叶片大小、边缘的形态在9株F_1代幼苗中与父本最相似,SG叶片为披针形,在9株F_1代幼苗中较为特殊。其余7株F_1代幼苗日渐成熟后再根据UPGMA聚类树状图进行形态多样性分析。
ISSR molecular markers were used to investigate the genetic diversity of 9 seedlings of the hybrid between Tiantina flower and Magnolia grandiflora. Nine F_1 generation seedlings were marked as SA, SB, SC, SD, SE, SF, SG, SH and SI according to their growth robustness. Ten ISSR primers were used to analyze the genomes of the parents and nine F_1 seedlings. The results showed that a total of 96 DNA bands were amplified in 11 plants, of which 85 were polymorphic, accounting for 88.5% of the total bands. UPGMA clustering results showed that all F1 generation seedlings were more similar to the hexaploid male parent than the diploid female parent. Among them, SC was the most similar to the male parent in 9 F 1 seedlings, and the SG was lanceolate in the shape of the edge, which was special among the 9 F 1 seedlings. The remaining seven F_1 seedlings matured according to UPGMA cluster analysis of morphological diversity.
